A Quantitative Description for Optical Mass Measurement of Single Biomolecules

چکیده
Label-free detection of single biomolecules in solution has been achieved using a variety experimental approaches over the past decade. Yet, our understanding magnitude optical contrast and its relationship with underlying atomic structure as well achievable measurement sensitivity precision remain poorly defined. Here, we use Fourier optics approach combined an structure-based molecular polarizability model to simulate mass photometry experiments from first principles. We find excellent agreement between several key experimentally determined parameters such contrast-to-mass conversion, accuracy, shape orientation dependence. This allows us determine mostly independent chosen, resulting general framework for light-based single-molecule quantification.

Mono-Size Distribution Index (MSDI): A new criterion for the quantitative description of size distribution
Graphical size distribution is widely used in different fields of science and studies related to powders, droplets, bubbles, and pores. However, in some condition it may also be necessary to express the size distribution quantitatively. In spite of there being several suggested ways to quantify size distribution in the literature, some of these approaches are not applicable for many methods and...
متن کاملQuantitative computer simulations of biomolecules: A snapshot
A recent workshop titled "Quantitative Computational Biophysics" at Florida State University provided an overview of the state of the art in quantitative modeling of biomolecular systems. The presentations covered a wide range of interrelated topics, including the development and validation of force fields, the modeling of protein-protein interactions, the sampling of conformational space, and ...
متن کاملA Quantitative Description of KcsA Gating II: Single-Channel Currents
The kinetic transitions of proton-activated WT KcsA and the noninactivating E71A mutant were studied at the single-channel level in purified, liposome-reconstituted preparations. Single-channel currents were recorded using patch-clamp techniques under nonstationary and steady-state conditions. Maximum-likelihood analyses reveal that the key influence of acidic pH is to increase the frequency of...
متن کاملQuantitative optical trapping of single gold nanorods.
We report a quantitative analysis of the forces acting on optically trapped single gold nanorods. Individual nanorods with diameters between 8 and 44 nm and aspect ratios between 1.7 and 5.6 were stably trapped in three dimensions using a laser wavelength exceeding their plasmon resonance wavelengths. The interaction between the electromagnetic field of an optical trap and a single gold nanorod...
متن کاملdevelopment of different optical methods for determination of glucose using cadmium telluride quantum dots and silver nanoparticles
a simple, rapid and low-cost scanner spectroscopy method for the glucose determination by utilizing glucose oxidase and cdte/tga quantum dots as chromoionophore has been described. the detection was based on the combination of the glucose enzymatic reaction and the quenching effect of h2o2 on the cdte quantum dots (qds) photoluminescence.in this study glucose was determined by utilizing glucose...
